Glycine itself acts as an inhibitory neurotransmitter, which provides a plausible mechanism, but marketing has run ahead of the data (see Sleep & Relaxation below).
What Is Magnesium Citrate?
FACT: Magnesium citrate is magnesium bound to citric acid. It is one of the better-studied organic magnesium salts, and at high doses it acts as a saline laxative — it is the active ingredient in FDA-regulated OTC laxative products (8).
Which Is Better Absorbed?
Short answer: Direct head-to-head evidence is lacking — neither form can currently be declared better absorbed than the other. Citrate has more published human absorption data; glycinate has been studied far less (2).

FACT: Magnesium citrate is a recognized osmotic laxative; FDA-regulated OTC products use it specifically for constipation at doses well above typical supplement doses (8). EVIDENCE: At supplement doses, looser stools are the most common reason people discontinue citrate (1, 8). UNCERTAINTY: The exact threshold varies widely between individuals.

FACT: A label that says “1,000 mg magnesium glycinate” refers to the compound, not the elemental magnesium. Elemental content is roughly 14% for glycinate and 16% for citrate — about 140 mg and 160 mg respectively per 1,000 mg compound. Always compare products by elemental magnesium per serving (1, 6).
FACT: General intake guidance is roughly 310–420 mg/day of magnesium from all sources for adults, depending on sex and age; the tolerable upper intake level for supplemental magnesium is 350 mg/day (1, 7). Total daily intake above the UL is the main risk factor for adverse effects.
